The Significance of DNA Content, Histological Grades, Cathepsin-D, and PCNA in Advanced Breast Cancers



Abstract
We investigated the possibility that DNA content, histological grades, Cathepsin-D and PCNA might have value as prognostic factors in breast cancer by studying paraffin-embedted tissue specimens We selected-randomly thirty patients of stage
III-breast
cancers who were diagnosed and treated between January, 1986 and December, 1988 and ten patients of benign breast disease for control. Breast cancer patients were divided into recurrent group (15 cases) and non recurrent group (15 cases) within 3
years
after surgery. We used paraffin embedded specimens for immunohistochemical staining using labelled streptavidin method and DNA content.
All benign and non-recurrent breast cancers were diploid whereas 6 cases were aneuploidy (40%) in recurrent breast cancer. Aneuploidy was more prevalent in metastazing more than 4 lymph nodes. Proliferative index were significantly higher in
breast
cancer than in benign diseases. But there was no difference between recurrent and non-recurrent groups. Aneuploidy was more frequently seen in cancers of high histologic grade than in low grade. Cathepsin-D had man value of 17.2% in recurred
group
and
14.9% in non-recurred group. There was no difference of Cathepsin-D positivity between recurrent and non-recurrent groups. The mean value of PCNA was 16.9% in recurred group and 8.9% in non-recurred group. the positivity of PCNA was higher in
recurrent
breast cancer than in benign lesion and non-recurrent breast cancer.
As a result of this study, it is concluded that DNA ploidy, histologic grade, and PCNA and thought to be useful proliferative factors in prdicting the recurrence. however, Cathepsin-D and proliferative fractions are not useful.
